Gaworski SUBJECT: Monthly Activity Report - October/November At your request I have prepared the following summary of my activities for the months of October and November, 1993. 1. The nitrate CSC skin painting study conducted at Microbiological Assoc. Inc. was completed in August. Unfortunately, the final report is still being prepared. M Wenk has indicated that a staff shortage is leading to a delay in the preparation of this report. His target date in January, 1994. 2. I have tried to encourage all of the biologists to complete some form of proposed work for the upcoming months. M. Dozier has prepared a separate proposal for "in-house" studies to be conducted next year with cellulose acetate fibers. Because of the scope of this work, a separate project number will be assigned and she will be responsible for completing the monthly status sheets. The proposal was completed in the TQM style which is apparently the style now preferred by J. Reid for cross-sectional projects. It was thought that this project could serve as a demonstration to the group. Since the project utilizes other groups in research, it will also give the group some visibility. A. Deaton has completed review of the genetic toxicity portion of the testing program. This review has lead to a proposed revision in the tests conducted. The review should be considered by SEAC. M. Bennett submitted a draft proposal for work in the area of cardiovascular toxicology. I thought that her ideas needed some clarification and she is continuing to look into this area for possible research. 3. Work on the cellulose acetate fiber project continued. A memorandum was prepared detailing the results of the 13 week exposure of the fibers to Gambles solution. This work resulted in other ideas regarding degradation of the these fibers which will be pursued as part of Mandy's proposal. 4. Inhalation studies are well underway at IITRI and will be completed in December. Previous problems experienced with the control boxes on the AMESA smoke machines have been resolved. We will continue to pursue the eventual change of this equipment to a computer controlled system after the first of the year. Almost daily contact has been maintained with the laboratory during this study. They have reported some problems with the animals turning in their holders because of small body size. In the future, larger animals will be obtained for the study. Filter shipments from the IITRI have been received on a routine basis and have been submitted to the smoking laboratory for nicotine and water analysis. A. Prakash was contacted regarding the possibility of analyzing serum samples for theobromine in rats exposed to D40 cigarette smoke. I managed to procure some rat serum for her to use to develop this analysis. Arrangements have been made with IITRI to supply the residual serum follow clinical pathology analysis at study termination. 5. The Sencar mouse skin painting study with the D40 cigarette has been initiated. There were some problems experienced during the initial collection of condensate by J. Bell. These appear to have been resolved. 6. A meeting was held with the biologists concerning the revision of the Masterbook for the Testing Program. Individuals will generate a generic protocol or protocol outline for the studies they monitor along with a brief justification for the inclusion of the study into a testing strategy. Once all of these are completed we can begin to revise them into a single document. 7. PACE reviews for M. Bennett and M. Dozier were completed. 8. Monitoring the activities of the biologists has continued. 9. General literature review in the area of inhalation toxicology continued. 10. The 1994 project request forms were completed for the testing program and the bioassay development program. The later program will account for the time spent in general laboratory activities. 11. The revised manuscript on the results of immunotoxicity testing has been returned to the editor for review. A letter was received on 18 Oct. acknowledging receipt of the revision, but we have not been notified of its final acceptance. 12. H. Ratajczak at IITRI prepard an abstract for the "Expeimental Biology 94" meeting in April, 1994. Heck, Dozier and Gaworski were listed as coauthors. The work describes the influence of gavage on systemic immunity and uses the data generated from our immunotoxicity screening studies.
