REYNOLDS SUBJECT: METHODS DEVELOPMENT FOR PLASMA NICOTINE To assess the possibility of using whole blood rather than plasma in Nicotine analysis, approximately 12 mls of blood must be collected. Mr. Riley Davis will be analyzing the Nicotine and Cotinine content on the upcoming 90 Day Levulinic Acid Study, and would like to determine if whole blood can be used for Nicotine analysis. The process would use whole blood from rats. The blood volume would be spiked then divided into two volumes, one plasma, the other whole blood. Both would be analyzed using gas chromatography. For this determination, approximately four to five rats need to be bleed. Dr. Kou-Mei Chang will be sacrificing six rats on Monday, June 12 and gave me permission to collect blood from these rats prior to sacrifice. These rats are controls and have not been treated chemically. The rats will be bled via retro-orbital sinus while anesthetized with 70 % CO2.
